Not at the moment - really tricky to mesh that with other query fields. Best bet is probably to create a new category, add all of the cards that meet your other search criteria (including the ones in the excluded category), then search for cards in the excluded category and remove them from the new category - the cards in that category will then be the ones you want.
 
Thanks for the tip, I did a Backup of the database, deleted the unwanted categories, and restored the db afterwards.
